First try his: go into your library folder (the one in your home folder) and find Mail>Mailboxes>Outbox, and toss it in the trash. The enxt time you start Mail, it will create a new one automatically. See if that works first.
Do you get any particular error messages? Check the activity monitor in Mail too to see if that gives any hints as to what's wrong.

This happened to a co-worker of mine. We solved the problem simply by quitting and relaunching Mail.
